The interaction of vitamin K1(VK1) with phospholipids was investigated using electrochemical, in situ UV-visible spectroelectrochemical methods, which showed that the electrostatic interaction between VK1 and lipids that revealed a rapid direct electron transfer process. In situ UV-visible spectra were obtained for the VK1 during the potential cycling from 0.8V to -0.8V. The absorption band appeared at 250 nm, and 360 nm. These bands are directly related to the redox reaction of VK1. The intermediate species of vitamin K1 was peculiarly stabilized on the PC coated GCE in buffer solutions. Anion radical absorption band appeared at 470 nm. The experimental parameters, such as pH, reducing potential and the stability of lipids layer are optimized to obtain best in situ UV-visible spectra. As the pH of the solution increased, the redox peak of modified vitamin K1 electrode shifted toward the negative potential.
