High throughput analysis of phosphatidylcholines from bovine brain using online capillary reverse-phase liquid chromatography coupled with LTQ-FT

Phosphatidylcholines(PC) from bovine brain were analyzed using online capillary reverse-phase liquid chromatography coupled with LTQ-FT equipped with home-built nano-electrospray ionization. cRPLC that has a maximum operation pressure of up to 10,000 psi has been shown to provide highest separation resolution observed to date in analyzing PC molecular species and thereby provides sensitive and efficient analyticcal method. Monoisotopic masses observed in a whole LC/MS experiment were clustered into unique mass classes (UMC). Out of a total of 2217 UMC, which correspond to different metabolic species, 75 PCs were identified to be known PCs from a single LC/MS experiment. The average mass measurement accuracy of the observed masses of PCs was calculated to be 5 ppm. The UMC method proposed in this study also provides information on relative quantitation for each PCs observed. This results shows that this experiment is a viable method for performing highly informative and high-throughput metabolite analysis, leading to discovery of potential biomarkers.
