A new capillary electrochromatographic (CEC) microchip with improved solution and chip handling, and reproducibility was developed. Patterned microchannels on a cyclic olefin copolymer(COC) substrate was packed fully with 0.8μm mono-disperse colloidal silica beads utilizing self-assembly packing technique followed by cover bonding by using a hot embossing machine. The chip was filled with buffer solution by self-priming capillary action. The packed silica at each channel served as a built-in nano-filter allowing quick and easy loading of samples and running buffer solution without filtration. Rreproducible control of electroosmotic flow was possible without leveling of the solutions in the reservoirs resulting 1.3% r.s.d. in migration rate because of large surface area-to-volume of the packed silica. The analytical characteristics of the chip were studied using FITC-derivatized amino acids as probe molecules. Mixtures of fluorescien isothiocyanate(FITC)-derivatized amino acids and FITC-derivatized peptides were successfully separated with less than 3mm separation channel length.
