Solid-phase synthesis of combinatorial libraries has emerged as a powerful tool for efficient drug discovery process. Recently, we have been exploring the potential of resin-bound anthranilic acid derivatives as versatile intermediates for generation of drug-like heterocyclic compound libraries and reported the solid-phase synthetic methods for 2-cyanoquinazolin-4(3H)-ones, 2,3-dihydrooxazolo[2,3-b]quinazolin-5-ones, 4-hydroxyquinolin-2(1H)-ones, 1,4-benzodiazepine-2,5-diones, and 2,1-benzothiazine-4-one-2,2-dioxides from the resins. On the other hand, we have introduced Irori X-kan system for split-pool combinatorial chemistry useful for highly efficient small organic molecule library generation as a part of our high-throughput synthesis platform development program. Herein we would like to present the first solid-phase synthetic method for construction of 2,1,3-benzothiadiazine-4-one-2,2-dioxide-based library through the resin-bound anthranilic acid derivatives, which could be efficiently adapted to Irori X-kan split-pool combinatorial chemistry system.
