Lab-on-a-chip technology is attracting great interest because the miniaturization of reaction systems offers practical informative advantages over classical bench-top chemical systems. Small volumes reduce the time taken to synthesize and analyze the product and reagent costs and the amount of chemical wastes can be greatly reduced. However, a highly sensitive detection method is required for monitoring the reaction process on a chip since the detection volume in the microfluidic channel is extremely small. Recently, highly sensitive optical detection methods for the fast and sensitive on-chip reaction monitoring have been developed in our research group. Various kinds of chemical/biological reactions, such as imine formation, DNA hybridization, and antibody-antigen immunoassay, have been performed in a specially designed PDMS microfluidic channel and their reaction processes were successfully monitored using the optical microscopic techniques. In this presentation, a fast and sensitive on-chip detection methods, based on confocal laser-induced microscopic technique, will be introduced.
