Electrochemical Enzyme Immunosensor with the Electrodeposited PVI-[Os(dme-bpy)2Cl]+/2+ Films on thiol-modified gold electrodes

We have been studied an amperometric immunosensor with the the electron-conducting redox polymer PVI-[Os(dme-bpy)2Cl]+/2+, a poly(N-vinylimidazole) (PVI) partially complexed with [Os(dme-bpy)2Cl]+/2+, (dme-bpy = 4,4’-dimethyl-2,2’-bipyridine), was electrodeposited on the thiol-modified gold electrodes. The assay comprised adsorption of rabbit lgG (r-lgG) and then Anti-rabbit lgG (Anti-r-lgG) and Anti-rabbit lgG-HRP (Anti-r-lgG-HRP) will compete for binding sites of r-lgG on the thiol-modified gold electrodes. Horesradish peroxidase (HRP) was used as a label of anti-rabbit lgG. In presence of H2O2, the Anti-r-lgG-labeling enzyme was electrically “wired” by the redox polymer film, converting the film into an electrocatalyst for the two-electron reduction of H2O2 to water at 0.0 V versus Ag/AgCl, the potential at which the H2O2 electroreduction current was monitored. The H2O2 electroreduction current decreased linearly with the concentration of the anti-rabbit lgG (Anti-r-lgG).
