Early diagnosis of OA is practically challenging and clinically important to provide patients better treatment. One of fundamental pathways in cartilage degradation is the significant up-regulation of MMP genes. Several lines of evidence suggest that matrix metalloproteinase-13 contributes to promote degradation of the cartilage. In the current study, we developed the MMP-13 active near-infrared fluorescence dark-quenched fluorogenic probe, Cy5.5-MMP13B3, for early diagnosis of OA in rat model in vivo. It was found that dark-quenched fluorogenic MMP-13 probe offers the potential of facile visual detection of MMP-13 in vitro and in vivo. In particular, by targeting biomarker in OA, symptoms and development stages of OA can be simply monitored, imaged, and analyzed in a rapid and efficient fashion. This platform technique can be widely applicable to any target proteases-based disease diagnosis and high-throughput drug screening by changing the specific peptide substrate linker between fluorophores and quenchers.
