The development of a gene delivery system with low toxicity, high delivery efficiency,and accurate release to target of interest is of paramount importance for gene therapy, treating diseases with specific genes. Although many viral and non-viral gene delivery systems have been developed to date,clinical use of gene delivery system is limited by toxicity (especially for viral systems) and low delivery efficiency(especially for non-viral systems).Here we used both gold nanoparticles(AuNPs) and lipid vesicles(liposomes)to improve the delivery of genes to interesting cell and thereby facilitate the use of genes for gene therapy. The results show that more cells are transfected when AuNP-liposome hybrids were used than when only cationic lipids or only AuNPs were used as gene delivery vehicles. Low cytotoxicity was also achieved with this approach because lower amount of cationic lipids was needed for effective transfection than when only cationic lipids were used. Interestingly, we observed two types of gene-modified AuNP-liposome hybrids-surrounded and encapsulated forms.
