Spatially resolved immobilization of biomolecules is essential to construct array-based biochips for diagnosis or drug discovery. Electrochemical patterning process on the preexisted electrode arrays is useful for device fabrication since each electrode can be individually controlled. We have developed a new electrochemical patterning method by using ITO electrodes array and random copolymer including an electrochemically active moiety. Polymeric monolayers are prepared on ITO electrodes. An electrochemical oxidation of the resulting polymeric monolayers enables thiol terminated molecules to be immobilized on the surface via thiol addition reaction. We use cyclic voltammetry, electrochemical impedance spectroscopy, infrared spectroscopy and fluorescence microscopy to show that the polymer modified ITO electrodes provide effective and selective surfaces for immobilizing alkanethiols. Furthermore, we demonstrate that aptamer-attached polymeric layers can be applied to detect proteins.
