There have been great demands for rapid, sensitive and versatile diagnostic tests for viral pathogens, warfare agents, cancer markers and other biomolecules for human welfare. Immunoassays are employed most widely in the field of medical diagnostics. This presentation describes the development and application of surface-enhanced Raman scattering (SERS) based immunoassay format. The probes named extrinsic Raman labels (ERLs) are prepared by the conjugation of Raman reporter molecule and antibody onto the surface of gold nanoparticles, which serve as detection labels in sandwich type immunoassay. ERLs not only provide means for ultrasensitive detection of various classes of biomolecules but also for the fundamental studies of SERS phenomena at nanostructure. By imparting magnetic property to the probe gold nanoparticle, the assay performance can be greatly improved. Examples of applications of both ERLs and magnetic ERLs will be presented.
