Enzyme immunoassay methods are widely used in the sensitive determination of special proteins or disease markers in clinical samples; they utilize enzyme-conjugated antigens (or antibodies) as signal generators for highly specific antibody-antigen immune reactions. A nonseparation electrochemical enzyme immunoassay (NEEIA) utilizes a microporous gold electrode. In this method, the amperometric signal is generated from the enzyme-labeled antibody bound to the electrode surface upon diffusion of substrate through the porous gold electrode. In an attempt to adapt the NEEIA approach to POCT measurements of C-reactive protein (CRP), one of the diagnostic markers for myocardial infarction, the use of gold electrodes formed on both sides of a porous nylon membrane is investigated: in this sensor design, one gold surface serves as a working electrode, and the other as a reference electrode. The CRP antibody was immobilized on the microporous gold electrode, and CRP-alkaline phosphatase was used as the enzyme conjugate.
