DREAM/ Calsenilin/ KChIP3, a multifunctional Ca2+-binding protein of the neuronal calcium sensor family, has been identified as a transcriptional repressor, a presenilin-interacting protein, and a Kv4 potassium channel-interacting protein. DREAM contains four Ca2+-binding EF-hands, and upon calcium stimulation, DREAM’s ability to bind to the DRE and its function as a transcriptional repressor are prevented. DREAM has been shown to play roles in several important neuronal processes, such as pain modulation and circadian rhythm. To better understand the role of DREAM in neuronal cells, we set up a strategy for the genome-wide identification of DREAM target genes. In our strategy, we first determine the consensus binding sequence of DREAM using SELEX technology. In parallel, we identify genes up-regulated upon perturbation using DREAM siRNA and caffeine-treatment using cDNA microarray analysis. Combination of two results and bioinformatic analysis discover novel DREAM target genes. Identification of DREAM target genes will help us understand the functional significance of DREAM and calcium signaling in neuronal cells.
