The spectrofluorimetric determination of hydrogen peroxide as an quenching reagent has been investigated by measuring the decrease of fluorescence intensity of tryptophan due to the complexation of hydrogen peroxide-tryptophan. An emission peak of tryptophan, which is decreased linearly by addition of hydrogen peroxide, occurs at 359 nm in aqueous solution with excitation at 288 nm. The determination of the hydrogen peroxide is in the range 1.0×10-7 M - 1.0×10-5 M and the detection limit is 3.0 × 10-8 M. The quenching effect of hydrogen peroxide on the fluorescence intensity of tryptophan may be considered on the basis of complexation between tryptophan and hydrogen peroxide and complies Stern-Volmer equation. The effects of foreign ions were investigated.
