Self-assembling nanostructures composed of DNA molecules have recently been demonstrated to be a highly useful material for constructing periodically patterned structures, mechanical devices with nanometer-scale feature resolution, and the unit structure of molecular computing systems with massive parallel fashion. Potential applications of DNA self-assembly and scaffolding include nanoelectronics, biosensors, and programmable/autonomous molecular machines. Here I present a mechanism of step-wise formation of DNA tile (4x4) by sticky-end association-dissociation via AFM analysis as a function of temperature and counterion concentration. The novelty of this structure includes a square aspect ratio with helix stacking and sticky-end connections in four directions within the lattice plane. Controlling the size of unit lattice has been achieved with slight reprogramming of the sticky-ends.
