In protein-drug delivery process, proteins may undergo physical denaturation and chemical degradation during microsphere fabrication. In this study, porous poly(lactide/glycolide) (PLGA) microspheres were fabricated by a emulsion-solvent evaporation technique using Pluronic F127 as an extractable porogen to prevent this denaturation and degradation. Then, lysozyme as a model protein was loaded by suspending microparticles into aqueous protein solution. Fabrication conditions such as PVA concentration, solvent ratio, stirring rate, PLGA/porogen F127 ratio have an important influence on particle size and microsphere pour size. The surface morphology of microspheres were investigated using scanning electron microscopy (SEM) for confirmation of its porous structure. Moreover, entrapment efficiency, release behavior from microspheres and biologically activity of protein were studied to evaluate the porous microspheres as a protein drug delivery system.
