An amperometric chloramphenicol (CAP) immunosensor was fabricated by convalently immobilizing an anti-CAT antibody on quantum dots modified-dendrimer bonded conducting polymer film. The immunosensor surface was characterized using QCM, impedance, XPS, and SEM techniques. The detection was based on competitive immuno-intereactions between free- and labeled-CAP for active sites of the anti-CAT by employing hydrazine as labels, which electrochemically catalyzed H2O2 reduction. The catalytic reduction was monitored amperometrically at -0.35V versus Ag/AgCl. Various experimental parameters, such as amount of anti-CAT, pH, temperature, applied potential, etc. were optimized and detection limit of CAP was determined. The proposed immunosensor was applied for analysis of CAP in real meat samples.
