We designed a system, which can detect Raman scattering and dynamic light scattering (DLS) at the same time. Raman spectroscopy is a powerful tool which provides molecular structure information with high sensitivity and molecular detection capabilities. DLS is uuseful technique for determination the size distribution profile of small particles in solution. Both Raman and DLS have been widely used in various areas. Using this simultaneous detecting system, we studied nanoparticles (NPs) aggregates on which molecules are adsorbed. Also, we applied to biological system. We prepared size controlled gold nanoparticles by sodium citrate reduction method. The NPs are functionalized with Raman encoding chemical and alkylbiotin. The biotinylated gold NPs are used to detect streptavidin. The specific binding of streptavidin with biotin leads to the cross-linking of particles. The cross-linking of NPs results in change of particles size and produces the Surface-Enhanced Raman Scattering (SERS) hot spots. We can detect binding event by DLS and specialty by SERS simultaneously.
