Biotinylation of Silica coated magnetic Nanoparticles and Their Conjugation with Avidin

Fe3O4 Superparamagnetic nanoparticles were synthesized by a chemical co-precipitation method and coated with biofunctional group(-NH2, COOH)and Biotin. Biotin, a small molecule, is well-known to bind tightly to the protein Avidin, with four biotins binding per protein(Kd = 10-15M). In this experiment, nano size Biotin coated magnetic particles on the silica based surface was prepared, which can efficienctly protect Fe3O4 core from chemical attack. The synthesized nanoparticles were characterized by FT-IR spectra, HR-TEM, EDS. Avidin binding Biotin coated Magnetic nanoparticles were analyzed MALDI-TOF-MS. In order to control magnetic nanoparticles , FITC-labelled Avidin was tagged on the Biotin coated particles and monitored fluorescence emission using a lab-built microscope equipped with CCD, TE-cooled PMT, and 488 nm diode laser. The Biotin coated magnetic nanoparticles will be applied to bio related study and biosensors.
