Photolithographic Fabrication of Micro-Patterned Prussian Blue (PB) Arrays for Electrocatalytic Analysis

A micro-patterned Prussian Blue (PB) array was fabricated using a soft photolithographic method for electrocatalytical detection of biomolecules. Photo-oxidation of alkylthiol SAM on gold by UV irradiation with a TEM mask generated a pattern, into which PB microarray was subsequently deposited. The microarray was characterized by cyclic voltammetry and optical microscopy. Compared to conventional PB film electrodes, PB arrays demonstrate much improved electrochemical stability, allowing hundreds of potential cyclings without noticeable deterioration of the film. The film thickness could be conveniently controlled by varying deposition time, yielding arrays of optimal film thickness that balances the stability and performance. Facile electron transfer was observed with redox probes and the effect of SAM insulation layer on response was investigated. The electrocatalytic analysis was demonstrated with biologically significant molecule ascorbic acid (AA). The calibration curve for AA shows a linear relationship over the concentration range of 1.0x10-5 to 8x10-7 M with a correlation coefficient of 0.99. The detection limit was determined to be 1 x10-8 M, which is 100 times lower than that on a conventional PB electrode. The method developed here provides a new surface for developing detection techniques for bioanalytes using electrocatalytical principles.
