DNA microarray technology is an increasingly important tool since it considerably accelerates genetic analysis. However, conventional microarrays have limitations in flexibility, speed, cost, and sensitivity. In addition to, most biochemical assays require secondary detection of a label. Here, we offer a new approach to study genotyping and gene expression profiling through Bio-AFM without any complicated labeling on the dendron modified surface. This novel method shows a sensitivity of ≤ 103 target DNAs detectable, a level that is better than the 105 number achievable with a high-density microarray system, and approaching the 103-104 level usually observed for quantitative PCR (qPCR) for genotyping study. The sensitivity of Bio AFM measurement was improved 105 times as microarray for gene expression profiling. The drawbacks that DNA microarray has poor detection and it is difficult for qPCR to detect biomolecules parallel can be overcome by Bio-AFM approach. We propose that lateral spacing of dendron modified surface can scale in a highly predictable manner (i.e., with dendron generation), force base AFM is also readily adaptable to other bio-chip systems.
