Establishment of efficient coupled assay system of microsomal prostaglandin E2 synthase-1 (mPGES-1) using purified 15-hydroxyprostaglandin dehydrogenase(15-PGDH) and mPGES-1

PGE2 is a member of prostaglandins that induces inflammation response, and mPGES-1 is responsible for the production of PGE2 in response to inflammation signaling. To set up an assay system of mPGES-1, we expressed mPGES-1 and 15-PGDH in E. coli and purified homogeneity. Then, a coupled assay system consisted of purified mPGES-1 and 15-PGDH was established. In this assay system, PGH2, the substrate of mPGES-1, is converted into PGE2, and PGE2 is oxidized to 15-keto PGE2 by 15-PGDH by the reduction of NAD+ into NADH. The amount of produced NADH was measured the fluorescence of NADH (ex = 340 nm, em = 468 nm) using a fluorescence plate reader. This assay system could be applied to screen chemical library to identify potent inhibitors of mPGES-1
