Osmoregulated periplasmic glucans (OPGs) are intrinsic components of the gram-negative bacterial envelope and are important for bacterial-host interactions. The OPGs of P. syringae are linear glucans having beta-1,2 and beta-1,6 linkages, together with those of Escherichia coli and Erwinia chrysanthemi. The glucans from P. syringae which are cultured in various conditions were isolated by trichloroacetic acid treatment and chromatographic techniques. Obtained compounds were characterized by thin-layer chromatography, matrix-assisted laser desorption/ionization-time of flight mass spectrometer (MALDI-TOF MS) and nuclear magnetic resonance (NMR) spectroscopy. The results demonstrate that depending on culture conditions, the degree of polymer is regulated and substituents such as succinyl or acetyl residue are added. Therefore, in this study, we report the appearance of synthesized OPGs is altered by culture conditions which are salt concentration, media type, incubated time and temperature.
