Structural study of human heat shock factor 1 using circular dichroism (CD) spectroscopy

Upon to heat stress, the elevated expression of heat shock genes such that Hsp70 or Hsp90 are rapidly synthesized and deployed to prevent protein misfolding and to assist in their refolding to the native state. Stress-induced transcription requires activation of heat shock factor (HSF). HSF1 is present in a latent state under normal condition, and it is activated upon heat stress by induction of trimerization and high-affinity binding to DNA. Here, we performed the circular dichroism (CD) spectroscopy to examine whether heat stress could influence the structure of HSF1. For this purpose, full-length HSF1 and its deletion mutations [including short-size HSF1 (1-120) and middle-size HSF1 (1-290)] were expressed in E.coli strains and purified by Ni column. As a result, structure of HSF1 could be influenced during the change of temperature. Especially, heat stimuli could change the structure of DNA-binding domain, but not unfold it.
