Structural study of heat shock factor 1 using fluorescence spectroscopy

All organisms have essential and evolved systems for responding to different stresses such as heat shock. In metazoan, the heat shock response is believed to be mainly regulated by the heat shock transcription factor 1 (HSF1). HSF1 is present in unstressed cells in an inactive monomeric form and becomes activated by heat and other stress stimuli. HSF1 activation involves trimerization and acquisition of a site-specific DNA-binding activity. HSF1 readily forms trimer in solution though interaction of its trimerization domain predicted to form a three-stranded coiled-coil. Here, we performed the fluorescence spectroscopy to examine whether heat stress could influence the structure of HSF1. For this purpose, full-length HSF1 and its point mutations (Trp23, Trp37, and Trp159) were expressed in E.coli strains and purified by Ni column. As a result, structure of HSF1 could be influenced during the change of temperature. Especially, Trp159 mutation protein show the similar results with wild-type HSF1, suggesting that the trimerization domain is very important in HSF1-activation.
