Structural characterization of platinum-binding BLM complex by NMR

Metallo-bleomycins recognize specific tumor cell, and noncovalently bind to DNA base pair to cleave sequence. In study, I refined BLM-A2 from BLM mixture. And an anticancer drug metal complex, platinum-binding BLM-A2 was synthesized by using microtitration in order to understand the function of metal complexes. Complete NMR signal assignment was accomplished by 2D NMR techniques which are gCOSY, TOCSY, NOESY, gHMQC, gHMBC. Comparison of chemical shift after complexation and NOESY experiment with different mixing times provided a detail structural feature of metal binding sites. Structural features of the complex were obtained by NMR-based distance geometry. 2D NOESY back-calculation method was performed to compare the obtained DG structure with experimental NOEs.
