Recently, we have reported a novel strategy to immobilize antibodies on solid surfaces by using a novel peptide conjugate, FcBP (Fc –binding peptide) which specifically bind to immunoglobulin G (IgG) Fc domain. As an extension of the research, we have fused FcBP with cell penetrating peptides (CPPs) to develop antibody transducer. CPPs have been employed for the cellular delivery of proteins, DNA or nanopaticles as their complexes, in order to test the cellular functions of the biomolecules or nanoparticles. There have been reported numbers of CPPs such penetratin, Arg8, HIV-1 TAT, MPS, etc. In this study, FcBP fusion peptides with various CPPs were constructed and tested for transduction of fluorescence-labeled Abs. When the Abs were monitored by fluorescence microscopy they were specifically delivered into living cells only in the presence of the fusion peptides. This represents the first peptide reagents, enabling Ab transduction into living cells without any covalent modification of antibodies, detergents, or electroporation.
