Subject Development of novel imaging agent for alzheimer’s disease using small peptide specific to amyloid beta

Abstract Amyloid-beta 42 (Aβ42) is produced from proteolytic cleavage of the amyloid precursor protein (APP) and it is accumulated forming the many amyloid structures which are the main cause of Alzheimer’s disease. For that reason, Aβ42 is known as a biomarker of AD. Here, we purified recombinant Aβ42 and confirmed the protein activity by making amyloid beta fibrils. Four different peptides (12mer) which bind to recombinant Aβ42 were screened via M13 phage display. The selected peptide shown micromolar range (μM) of binding affinity to target Aβ42. We verify the binding ability of peptide using immunoprecipitation-like methods immunofluorescence methods. Our protein is well precipitated using screened peptides. And high percent (>80%) of merging image were observed when the brain tissue (3X Tg-mouse) stained using peptide and antibody. Finally, to design a peptide based contrast agent, we conjugated Aβ42 specific peptides to the ultrasmall superparamagnetic iron oxide (10nm) through EDC/NHS coupling reaction. Such peptide based USPIOs that target neurotoxic accumulation of Aβ42 are potentially useful for early diagnosis of Alzheimer’s diseases and disease management.
