Subject Cationic Chlorin and Polyoxometalate Nanocarrier Synthesized by Click Chemistry for Nuclear Targeting and Highly Efficient Photodynamic Therapy In Vitro

Abstract Simple click chemistry enabled the synthesis of neutral clicked chlorin, which was converted to cationic clicked chlorin that formed a clicked chlorin-polyoxometalate (POM) supramolecular complex as a nanocarrier via electrostatic interactions between the cationic clicked chlorin and the polyanionic POM. The cellular uptakes of clicked chlorin, cationic clicked chlorin, and the cationic clicked chlorin-POM complex against A549 cell lines were confirmed by confocal laser scanning microscopy. Clicked chlorin, cationic clicked chlorin, and the cationic clicked chlorin-POM complex were accumulated in cytoplasm as well as nuclei. Cell viability results showed that the cationic clicked chlorin-POM complex (IC50, 338.8 nM) and free cationic clicked chlorin (IC50, 342.4 nM) exhibited significantly enhanced, more than 10 times higher, photocytotoxicity compared to clicked chlorin (neutral, IC50, 3.81 μM) based on better nuclear targeting. This results suggest that this cationic clicked chlorin-POM complex is a highly promising photosensitizer for PDT without aggregation and dark cytotoxicity for no side effect.
