Subject Effect of ERLBD302-552 on the amyloid fibril formation of α-synuclein

Abstract α-Synuclein (αSyn) protein of Parkinson’s disease patient undergoes conformational change to form amyloid fibrils as insoluble deposit. In our previous study, we have constructed the plasmid ERLBD302-552/pET28 ans overexpressed the His-tagged ERLBD302-552 protein by IPTG induction using BL21(DE3). In the present study, we examined the process of amyloid fibril formation of αSyn in the presence of His-tagged ERLBD302-552 in vitro. We also made the fusion construct ERLBD-L23-αSyn/pET28a by cassette ligation. Finally we discuss the function of ERLBD302-552 on the amyloid fibril formation of αSyn based on the ThT fluorescence kinetics of fusion protein.
