Abstract Although numerous kinases have been reported to regulate HIF-1α stability and activity, few phosphatases are known to be involved in HIF-1α regulation. In this study, we report that ADP-ribose (ADPR) pyrophosphatase, nudix hydrolase 9 (NUDT9) negatively regulates HIF-1α accumulation by modulating AMP-activated protein kinase (AMPK) signaling pathway. Knockdown of NUDT9 activates transient receptor potential cation channel subfamily M Member 2 (TRPM2), resulting in Ca2+ influx mediated activation of calcium/calmodulin-dependent protein kinase kinase (CAMKKβ). AMPK activation by CAMKKβ is shown to enhance HIF-1α accumulation and the possible downstream effector for AMPK is identified. Our finding provides insights into the roles of NUDT9 phosphatase as additional, discrete modulator of hypoxia stimulated HIF-1α accumulation through different signaling pathways.
