Subject Hetero-spectral 2D correlation analysis of the denaturation process of α-lactalbumin by fluorescence and IR spectroscopies

Abstract It is important to understand the denaturation mechanism of protein because it provides fundamental knowledge in the study of disease caused by protein denaturation. A representative whey protein, α-lactalbumin undergoes an intermediate state at a specific pH during denaturation process. In this study, the pH-induced denaturation process of α-lactalbumin was explored by fluorescence and IR spectroscopies. Principal component analysis (PCA) and hetero-spectral two-dimensional correlation spectroscopy (2DCOS) were used to understand denaturation mechanism of α-lactalbumin. Details on the results of pH-induced fluorescence and IR spectral changes of α-lactalbumin will be discussed in this presentation.
