Subject Hypersensitive Cancer Immunosensor Based on One-Shot Dual Signal Detection on Nano-patterned Biochip

Abstract Three-dimensional enhanced dark-field super-resolution microscopy (3D EDF-SRM) based on hypersensitive one-shot dual signal detection was developed for immunoassay of TNF-a as a cancer biomarker. The 3D EDF-SRM is equipped with a dual view and a digital single-reflex (DSLR) camera, enabling dual signal detection which was both quantifiable and qualitatively. The standard TNF-a was quantitatively analyzed by measuring the scattering signals of 80 nm silver nanotag on an array chip with 100 nm gold spot. The localization precision in nanotag on the gold spot narrowed to <10 nm after applying the least-cubic algorithm. The developed hypersensitive immunosensor exhibited a detection limit of zeptomolar level (S/N = 3) with a wide dynamic detection range of zM-pM. In addition, a recovery greater than 98% was achieved by spiking standard TNF-a into human serum samples.
