Subject Synthesis of EGCG-amide and its specific cytotoxicity against highly metastatic breast cancer (MDA-MB-231) cells

Abstract (-)-Epigallocatechin gallate (EGCG), a major green tea flavonoid, is well known for its cytotoxic activity against various cancer cell lines. In this study, a new EGCG derivative with an amide linkage at the C3 position was synthesized by using oxidation followed by reductive amination strategy of the globally protected epigallocatechin. The newly synthesized EGCG-amide was investigated for its anticancer activity against a panel of cancer cell lines to show strong cytotoxicity against the highly metastatic breast cancer cells (MDA-MB-231). Given the importance of metastasis in the cancer treatment, the mechanism for specific cytotoxicity of the EGCG-amide against the MDA-MB-231 cell lines is of particular interest. Synthesis, anticancer activity, and identification of the target proteins of EGCG-amide will be presented.
