Subject Solid-Phase Parallel Synthesis of 1,3-thiazole Derivatives Based on Peptidomimetics

Abstract Synthesis of peptidomimetics has known to be effective for better understanding of protein aggregation which causes neurodegenerative diseases such as Alzheimer’s disease. In this work, solid-phase synthesis of 1,3-thiazole derivatives based on peptidomimetics is described. The synthetic strategy starts from constructing 1,3-thiazole core skeleton resin by cyclization of resin-bound cyanocarboimidodithioic acid with ethyl bromoacetate. The resulting core skeleton has amino group and carboxylic acid group which subsequently undergoes functionalization with various amino acids by amide coupling. The desired product has core skeleton of 1,3-thiazole with two sets of three successive amino acids. Trifluoroacetic acid (TFA) in dichloromethane (DCM) is used for cleaving the resin.
