Whole saliva is complex biomolecules containing such as a variety of mucin, proteins, DNA, RNA, hormones and electrolytes. Glycoproteins are major of proteins such as α-amylase, mucin, immunoglobulin, lactoperoxidase and lactoferrin in saliva fluid and there are well known as key factor of immune response. A glycosylation change of saliva could reflect the condition of health because glycosylation is highly sensitive to the biochemical environment and it has been implicated in many diseases. However the salivary glycome analysis has not been well studied and were significantly hindered by the inherent component of saliva. Here, we aim to develop the method for more selective and efficient enrichment of glycans directly released from whole saliva. Human saliva were collected from 20 individuals for exploring the salivary glycome. To efficiently eliminate interferences such as electrolytes and small biomolecules, saliva was filtered by 10K MWCO. The supernatants were digested with PNGase F to release N-glycans, then were fractionated according to size and polarity by graphitized carbon solid phase extraction. Enriched glycans were analyzed by MALDI-TOF/TOF MS. We could successfully enrich the N-glycans through pre-filtering procedure in MS analysis, especially, acidic glycan fractions.
