Saliva, which is secreted mainly from parotid, submandibular, and sublingual glands, contributes to many biological roles and functions in mouth. Saliva is constituted a highly complex mixture such as water, electrolytes, proteins, lipids, and carbohydrates. Glycoproteins are also one of the major components in saliva. Those are involved in the maintenance and protection of oral health against decay, damage, and microbial inflammation. Although studies of profiling of saliva glycoproteins and glycans have been reported, saliva glycosylation has not been well characterized due to inherent complexity and diversity of glycome. In this study, salivary N-glycans have been intensively characterized including not only the compositional and quantitative analysis by nano-LC/MS but also structure elucidation using tandem MS. N-glycans were enzymatically released from saliva after enriching high molecular weight compounds by centrifugation. Solid phase extraction with a porous graphitized carbon cartridge was used to purify and fractionate only N-glycans. We profiled N-glycans using nano LC-chip/Q-TOF MS. Interestingly, we found highly fucosylated glycans (Hex3-7HexNAc3-7Fuc1-7) uniquely present in saliva in contrast with other human fluids such as serum and milk. The structures of novel N-glycans containing 3 to 6 fucoses on outer position were elucidated for the first time by diagnostic fragment ions and fragmentation patterns during tandem MS. Saliva N-glycome may be applied to biomarker discovery of diseases and post-mortem interval estimation in forensic science.
