Buckminsterfulleren C60 is one of famous nanoparticle with various biological activities. It possesses several interesting properties and can be used in medicine and nanotechnology. However, it also has general toxicity and adverse effects that are poorly understood. To understand both effects of C60, we have developed specific RNA aptamer to recognize C60-fullerene. The SELEX (Systematic Evolution of Ligands by EXponential enrichment) procedures were carried out to obtain RNA aptamer. After 9 cycles of selection, pull-down assay showed that a pool of C60-specific RNA was enriched. The enriched aptamers were cloned to afford several different sequences. One of these clones displayed low micromole binding affinity to C60 fullerene and did not bind at all to C70 fullerene. Results show that RNA aptamers could be used as tools to detect a typical nanoparticle, C60, both in biological and environmental system.
