Fluorescence imaging in particular is well-suited for live imaging of biological Zn(II), which has no facile spectroscopic or magnetic signature. Successful application of this methodology requires the development of robust Zn(II) imaging agents with high sensitivity, selectivity, and temporal fidelity. A N2O2-type dipyrrin based bimolecular zinc(II) complex has been synthesized and shown to have a sharper, blue-shifted, and more enhanced fluorescence emission. About three-fold fluorescence enhancement was achieved within micromolar concentration which is an important parameter for Zn(II) detection in vivo. The increase of emission intensity is due to the dominant role of aryl-ring rotation in governing the excited state dynamics and fluorescence properties of dipyrrin dye. The fluorescence titration revealed that the metallo ligand showed a very strong zinc(II) binding affinity than second zinc(II) binding in dimeric chloro-bridged complex. The fluorescence emission changes of dipyrrin dye to zinc can be viewable not only by instrument but also by naked-eye (violet → sky blue).
