High-efficiency separation techniques, such as capillary electrophoresis (CE), coupled to a mass spectrometer offer the ability to separate and identify chemical information on analytes in small sample volumes. However, the salts and detergents used in the mobile phase for CE separations suppress ionization efficiencies and contaminate the inlet of the mass spectrometer. This study describes a new method that uses desorption electrospray ionization (DESI) to overcome these limitations. Effluent from capillary columns is deposited on a moving heating plate that is heated with electric heating coil. As the surface moves, the temporal separation of the eluting analytes is spatially encoded on the surface. Then, using DESI, surface-deposited analytes are preferentially ionized, reducing the effects of ion suppression and inlet contamination on signal and then several important experimental parameters influencing electrospray ionization of sample were studied. With the use of this novel approach based separations were performed on several glycoproteins such as fetuin in order to validate this method.
