Photoreceptor proteins play crucial roles in receiving light stimuli and transmitting the light-generated signal to their signal partners. However it still remains challenge to capture the detailed time-dependent change of the three-dimensional structures along their functioning in their native aqueous environment, even for photoactive yellow protein (PYP), the model system that has been extensively studied by various methods. Here we employ pump-probe X-ray solution scattering to track detailed structural transitions during the photocycle of PYP after excitation at 460 nm in a wide time range from 3.16 us to 300 ms and capture four structurally distinct intermediates and their associated five time constants (10 μs, 280 μs, 1.3 ms, 23 ms, and 650 ms).
