In this study, magnetic bead with specific oligo-nucleotide sequence was synthesized so that it can be used to capture DNA or oligo-nucleotide. Magnetic bead with COOH functional group was purchased. The magnetic beads were reacted with L2-(N-morpholino)-ethane sulfonic acid to produce amide bond and incubated with vortexing. To check and monitor the reaction process, Fourier transform infrared spectroscopy was employed. NH2 modified oligo-nucleotide was added to react with the modified magnetic bead. The magnetic beads attached with oligo-nucleotide were reacted oligo-nucleotide with matched sequence. The hybridization reaction between functionalized beads and target oligo-nucleotide was performed in hybridization buffer at room temperature. The hybridized beads were washed and resuspened in ultra pure H2O. Capture oligo-nucleotide was released by heating and then analyzed by MALDI mass spectrometry.
