The determination of the content of nicotinamide and nicotinic acid is performed by reversed phase high performance liquid chromatography with UV detector. The performances of ODS C18 column which are frequently used in the analysis were compared using two standard samples (Nicotinamide and nicotinic acid) and five real samples. Isocratic elution was used to elute out sample using sodium dihydrogen phosphate buffer and acetonitrile (V/V=90/10). The buffer solution was prepared by weighting 0.78 g sodium dihydrogen phosphate, 0.78 g sodium 1-heptanesulfonate and 0.35 mL phosphoric acid into a 900 mL dissolved with deionized water. The absorbance of the samples was detected at 250 nm and 260 nm. The UV detector wavelength was using of 250 nm and 260nm. Retention time, adjusted retention time, intensity, W1/2, number of plates, and capacity factors were calculated based on the LC chromatograms. nicotinic acid (~3.12 min) eluted a little earlier than nicotinamide (~3.91 min).
