Glycosylation is one of the most common post-translational modifications in proteins, with 70% of human serum proteins being glycosylated. Carbohydrate structures have been shown to change with various pathological states including cancer. The immense structural and linkage heterogeneity of glycan structures makes it critical to fully characterize the glycome, including isomers, as a way to better understand their biological function. Currently there is no method for high throughput structural elucidation, but significant progress has been made in the determination of structures using a combination of methods including exoglycosidase digestion and tandem mass spectrometry. Separation problems have all largely been solved using HPLC or nanoLC and graphitized carbon. The next level of analysis is to determine structures rapidly. For this purpose, we are developing a library of glycan structure that can be used to identify structures rapidly. We have determined that the number of glycan structures in many biological systems are relatively small and can be thoroughly characterized. By building a library of structures with HPLC retention time, accurate mass, and tandem MS, we can identify glycans with very low false positive rate. We use this library to rapidly identify structures from complex unknown N-glycan mixtures like serum, milk, or saliva.
