Mitochondrion is known as a power-house of human body, where ATP generation is carried out and biological energy is produced. Thanks to the specific topology of inner membrane, that is called folding structure, more surface area is available not only for ATP synthesis but also for many cellular events which are taken place on it. Three major components of lipids composing of the mitochondrial inner membrane are phosphatidylcholine (PC, 40% in weight), phosphatidylethanolamine (PE, 40% in weight) and cardiolipin (CL, 20% in weight). The last one, CL, is a complex phospholipids only found in the energy-transducing membranes of bacteria and mitochondria. It is considered as an essential component in maintaining stability and carrying out the biological functionalities. Therefore, our interest has been decided to address on the nature of folding structure and whether or not CLs are involved. According to the physics viewpoint, because of the inverted cone shape of cardiolipin, it tends to form a spontaneous curvature in case of monolayer and non-lamellar phase in case of bilayer membrane. On the other words, cardiolipin might play a key role for making the folding structure in inner mitochondrial membrane. We use the Langmuir-Blodgett monolayer technique to characterize the phase behaviour and the elasticity of monolayer system, which is composed of CLs and DPPCs, and prove the contribution of CLs in forming the folding structure on liquid substrate, using the in-situ Langmuir monolayer-X-ray reflectivity combination. The folding insight has been modelled in term of electron density, roughness and thickness, respectively in order, of lipid tail-region.
