A simple and direct method for diagnosis of Salmonella typhimurium in urine sample is developed. An electrochemical genosensor based on selective target gene is fabricated by immobilizing probe DNA onto the nanostructured conducting polymer film. The sensor probe was characterized using XPS and SEM. Fresh bacterial isolates were cultured and enumerated on to the nutrient agar media and then diluted in filtered urine sample. All bacterial cell solutions were sonicated and further treated with phenol, isoamyl alcohol solution. The treated bacterial lysate was incubated with the fabricated sensor probe. Electrochemical detection to detect target gene in the cell lysate was conducted using cyclic voltammetry and impedance spectroscopy. Results obtained shows increased in the signal response when samples were treated with phenol, isoamyl alcohol solution. The dynamic range for S. typhimurium detection was between 102 cells ml?1 and 107 cells ml?1 with a low detection limit of approximately 74 cells ml?1 (RSD <5%). The developed genosensor exhibits high selectivity toward Salmonella typhimurium and shows negative response for other gram negative bacteria (GNBs) including Shigella soni and Escherichia coli.
