The study of structural transition dynamics of proteins is essential for understanding the structure-function Relationship. Thus far, the study of protein structure using pump-probe time-resolved X-ray solution scattering has recently evolved. However, since rotationally averaged scattering arises from randomly oriented sample molecules in solution, this gives the scattering pattern a reduced dimension. Determining the exact structure of a protein from X-ray solution scattering data always suffers from lack of information. To circumvent this obstacle, we have developed a method that can help us increase the structural information content of the solution scattering signal: heavy-atom labeling scheme. In this scheme, the target residue sites in the protein are labeled with heavy atoms by using well-established labeling scheme. We could substantially increase the information content of the protein structure in a site-selective manner. Here, we discuss on the time-resolved X-ray solution scattering signal of heavy-atom labeled myoglobin. The time-resolved X-ray solution scattering with heavy-atom labeling technique is a powerful means for study of structural transition dynamics of proteins in solution.
