A silica-coated magnetic nanoparticles (Si-MNPs) functionalized with thiol (-SH), disulfide (-S-S-), carbon chain (-C18), carboxyl (-COOH), amine (-NH2), and aldehyde (-CHO) group have been designed and constructed that the capacity of effective binary proteins, which are bovine serum albumin (BSA) and lysozyme (LYZ) separation from mixed-state solution. The hydrophobic functional groups such as thiol, disulfide, and carbon chain induced protein adsorption onto the Si-MNPs surface than other hydrophilic functional groups under the specific conditions as at pH 4.65 for BSA and pH 11 for LYZ. However, in mixed-state proteins, the driving force of BSA and LYZ to the Si-MNPs was highly affected by their molecular weight rather than optimal pH value. Significantly, it is expected that this findings can be extended to various biomolecules separation from intact biological samples consisting of a plurality of proteins.
