Reactive oxygen species (ROS) such as hydroxyl radical, superoxide and hydrogen peroxide have been shown to be involved in aging. The direct scavenging effect of spermidine on DPPH radical, H2O2 and DNA oxidation related to oxidative stress were evaluated in-vitro. In addition, its inhibitory effect on the activity and expression of matrix metalloproteinase-2 related to metastasis were determined using gelatin zymography in human dermal fibroblasts (HDFs). It was observed that spermidine exhibited scavenging activity on DPPH radical above 500 ?M. Especially, spermidine at 500 ?M showed scavenging activity of H2O2 and DNA oxidation. Furthermore, the expression level of antioxidant enzyme such as superoxide dismutase was increased in the presence of spermidine. Gelatin zymography assay showed that spermidine inhibited MMP-2 activity. In addition, the expression level of histone deacetylase gene was increased in the presence of spermidine in RT-PCR assay. This results indicate that ROS is decreased by treatment of spermidine via up-regulated histone deacetylase. We suggest that spermidine can use as a potential agent to prevent ROS-related diseases including inflammation, cancer and aging
