Recently, due to the mass production and widespread use of manufactured nanomaterials (MNs), there are increasing concerns on the potential hazards of MNs. However, many limitations and challenges of the current nanotoxicity assessment protocols were often found and accumulated in the literature. In this study, we have developed and proposed a novel flow cytometry protocol to distinguish intrinsic NP toxicities directly associated with the uptake of NPs. Particularly, NP associated ROS generation for the HeLa cells exposed to positively and negatively charged ZnO and SiO2 NPs were investigated. The results shown in this presentation have demonstrated the feasibility of assessing the intrinsic effects of various physicochemical properties of NPs (e.g., hydrodynamic sizes, surface charge, and solubility of NPs) on the cellular responses related to various cell death mechanisms (e.g., apoptotic and necrotic cell death).
