Combinatorial chemistry has become an extremely powerful technique for the generation of small, drug-like, organic molecule libraries and for medicinal chemistry programs and drug discovery processes as a key objective within the pharmaceutical industry. In combinatorial synthesis, solid-phase organic synthesis (SPOS) is now routinely used to prepare a large number of small, heterocyclic, drug-like molecules and is especially useful in creating massive numbers of hit and lead compounds as part of high-throughput screening technologies. As part of an ongoing drug discovery project, we required concise solid- or solution-phase routes for the construction of a novel drug-like chemical library. We have developed an efficient, facile, and rapid solid-phase synthetic strategy for construction of triazoloamides and three types of fused-thiazoles (thiazolothiazine, thiazolopyrimidine diones, thiazolopyridine)
